Das Prinzip des TPHA-Tests

NewBio TPHA ist ein passiver Partikelagglutinationstest für den qualitativen Nachweis von IgG- und IgM-Antikörpern gegen Treponema pallidum in Humanserum, EDTA-Plasma oder Liquor und zur Bestimmung des Titerwerts der Proben.

NewBio TPHA verwendet konservierte Geflügel-Erythrozyten, die mit extrahierten Antigenen von T. pallidum (Nichols-Stamm) beschichtet sind. Spezifische Antikörper, die in einer Plasma- oder Serumprobe vorhanden sind, binden an diese Antigene, wenn die Probe mit den Partikeln inkubiert wird. Dadurch agglutinieren die Partikel, setzen sich ab und bilden ein charakteristisches Muster in der Testvertiefung.

Unspezifische Reaktionen werden durch den Einsatz von Absorptionsmitteln eliminiert.

Eine Studie mit 1248 syphilisnegativen EDTA-Plasmaproben ergab eine Spezifität von 99,92 % (95 % Konfidenzintervall 99,55-100,0 %).

Sensitivität

Eine Studie mit 205 Syphilis-positiven Proben (48 EDTA-Plasma und 157 Seren) ergab eine Sensitivität von 100 % (95 % Konfidenzintervall 98,22-100,0 %).

Nachweisgrenze

NewBio TPHA hat eine erwartete Nachweisgrenze von ≤0,1 IU/mL gegenüber dem WHO 1st IS for human syphilitic plasma IgG NIBSC code: 05/122.
